How the assay works
In Neutrophil Elastase Inhibitor Assay Kit protocol, Neutrophil Elastase hydrolyzes a specific fluorescent substrate to release the fluorescent group, which can be detected at Ex/Em = 400/505 nm. In presence of a potent neutrophil elastase inhibitor, the hydrolyzation of substrate will be inhibited or stopped. For comparison of the relative efficacy of test inhibitors, a control inhibitor, SPCK (Ki = 10 µM for human leukocyte elastase) is included.

- Test inhibitor preparation Neutrophil elastase preparation
- Incubate for 5 mins at 37°C
- Add substrate mix
- Measure fluorescence (Ex/Em = 400/505 nm) in a kinetic mode
- Incubate for 30 mins at 37°C protected from light
- Measure fluorescence (Ex/Em = 400/505 nm) in a kinetic mode

This product is manufactured by BioVision, an Abcam company and was previously called K782 Neutrophil Elastase Inhibitor Screening Kit (Fluorometric). K782-100 is the same size as the 100 test size of ab118971.
